Hemoglobin electrophoresis is a well-established technique routinely used in clinical laboratories to screen patient samples for hemoglobinopathies (hemoglobin variants and thalassemias).
Many international societies such as TIF(1), CDC and APHL(2) recognise the benefits of the Sebia Capillary Electrophoresis (CE) technology, such as the high resolution and its clear hemoglobin fractions profile in the screening of hemoglobin disorders.
In addition to the Capillary electrophoresis, Sebia provides Gel technique-based solutions: HYDRAGEL HEMOGLOBIN(E) and HYDRAGEL ACID(E) HEMOGLOBIN(E) that enable, on the HYDRASYS instruments the separation of the hemoglobins and the detection of the major hemoglobin variants: S or D and C or E, by electrophoresis on alkaline agarose gels.
Electrophoresis on acidic gels, like HYDRAGEL ACID(E) HEMOGLOBIN(E), should follow to confirm the identification of hemoglobin variants, mainly, to differentiate hemoglobins S from D and E from C.
The visualization of the gel provides qualitative and quantitative (HbA and HbA2) analysis.
CAPILLARYS instruments perform all sequences automatically, from primary tube sampling to final clear-cut profile, with precise quantification and presumptive identification of the most common hemoglobins.
CAPILLARYS instruments allow high resolution separation of the major hemoglobin variants (Hb S, Hb C, Hb D and Hb E) and accurate quantification of the Hb A2 and Hb F.
Furthermore, the Sebia CE technique is highly reproducible and the migration position given by the software can be used as an aid in the identification of Hb variants(3).
Capillary zone electrophoresis allows clean separation of Hb E from Hb A2 and facilitates easier detection of Hb Bart’s and Hb H, the X-axis migration position is a great help in the presumptive identification of hemoglobin variants(3).
Thanks to the broad range of capillary instruments, the hemoglobin technique answers the needs and variable workload of all laboratories involved in abnormal hemoglobin screening.
Whole blood sample are used and are analysed directly on Capillary instruments.

In this report, we evaluated the use of the capillary electrophoresis (CE) migration position in the CAPILLARYS 2 CE instrument.
This article assesses the performances of the different manufacturers and technologies (HPLC and CE) for Hb A2 measurement using both International Reference Reagent for HbA2 (WHO IRR 89/666) and analysis of three whole blood specimens over a range of HbA2, distributed to 56 laboratories located in 14 different countries.
